Aziz Sancar was born in Savur, Mardin, Turkey. He received a M.D. degree in 1969 from Istanbul University School of Medicine and for a period practiced medicine as a village doctor in Turkey. He received a Ph.D. degree in Molecular Biology in 1977 from the University of Texas at Dallas where he worked on DNA photolyase under the guidance of the discoverer of photolyase, Claud S. Rupert. Sancar did his postdoctoral work at Yale University Department of Molecular Biophysics and Biochemistry where he studied nucleotide excision repair. In 1982, he was appointed associate professor in the Department of Biochemistry, University of North Carolina, Chapel Hill. He became full professor in 1988 and Sarah Graham Kenan Professor in 1997. Sancar's work is on DNA repair. He invented the maxicell method for identifying plasmid encoded proteins and this method has become a standard tool in molecular biology. Sancar reconstituted the 3-subunit bacterial and the 6-subunit human DNA repair excision nucleases and discovered that DNA lesions are removed by a dual incision mechanism that excises 12-13 nucleotide oligomers in bacteria and 24-32 nucleotide oligomers in humans. He discovered the transcription repair coupling factor which couples these two processes in bacteria and found that the human general transcription factor also functions as a general repair factor. Sancar has worked for 25 years on the light-activated DNA repair enzyme, photolyase. Using genetic, molecular biological, physical-chemical, and crystallographic methods he has elucidated the reaction mechanism of the enzyme which involves intramolecular energy transfer between two chromophores and photoinduced intermolecular electron transfers between enzyme and substrate. Sancar's work on photolyase has led his group to discover the second photosensory pigment in the human eye which is called cryptochrome and which regulates the biological clock. Sancar received the Presidential Young Investigator Award in 1985, the American Society for Photobiology Research Award in 1990, and the Basic Science Research Award from the Turkish Scientific Research Council in 1995. He was elected an Associate Fellow of the Third World Academy of Science in 1996.
